Dr. Sack’s research interest aims to understand the mechanisms regulating mitochondrial metabolism and the role of this organelle in the pathophysiology of cardio-metabolic syndromes. His laboratory focuses on multiple levels of mitochondrial regulation and employs functional genomic modulations to investigate how perturbations in mitochondrial biology effect cardiac stress-tolerance and modulates peripheral tissue insulin and glucose sensitivity. A core component of his laboratories work is to additionally translate these findings to the clinic to establish whether the regulatory programs operational in murine models translate to human pathophysiology. In his effort to elucidate regulatory controls of the mitochondria in governing stress tolerance, Dr. Sack’s laboratory focusses on post-translational control of mitochondrial function with specific emphasis on acetylation and succinylation with the major focus on the rgulatory roles of Sirt3 and GCN5L1. In clinical studies his laboratory employs fasting and refeeding as a model system to modulate mitochondrial protein function and assay their effect on human biology. His current translational work explores the role of mitochondrial control of innate and adaptive immunity.
